: In order to study the role of prostaglandin in the regulation of the gastrointestinal functions, gene expression of prostaglandin receptors along the rat gastrointestinal tracts were investigated.
: Rats were used for the study. The combination of counterflow elutriation separation of mucosal cells and Northern blot analysis was used to detect the gene expression of prostaglandin receptors in gastrointestinal tracts.
: In small intestine and colon, prostaglandin E2 EP1 and EP3 receptor mRNAs were mainly localized in the deeper intestinal wall containing muscle layers. EP4 receptor gene expression, on the other hand, was detected in the intestinal mucosal layer.
In the stomach, EP1 mRNA was detected in gastric muscle layers, whereas EP3 and EN receptor gene expression was mainly present in the gastric mucosal layer containing epithelial cells. In gastric epithelial cells, parietal cells were found to have both EP3 and EP4 receptors. At lower concentrations, prostaglandin E2 inhibited gastric acid secretion by parietal cells probably through EP4 receptors. At higher concentrations, however, it stimulated it. On the other hand, mucous cells possessed only EP4 receptor mRNA.
: Thus, it is suggested that prostaglandin E2 modulates gastrointestinal functions through at least three different prostaglandin receptors (EP1, EP3, and EP4), each of which has a distinct distribution in the gastrointestinal tract. 相似文献
Regenerating islet-derived protein (Reg)3β belongs to a member of the Reg family of proteins and has pleiotropic functions, including antimicrobial activity and tissue repair. However, whether Reg3β plays a protective role in the development of colitis and ileitis has not been fully investigated. We generated transgenic mice expressing a short form of cellular FLICE-inhibitory protein (cFLIPs) that promotes necroptosis, a regulated form of cell death. cFLIPs transgenic (CFLARs Tg) mice develop severe ileitis in utero. Although Reg3β is undetectable in the small intestine of wild-type embryos, its expression is aberrantly elevated in the small intestine of CFLARs Tg embryos. To test whether elevated Reg3β attenuates or exacerbates ileitis in CFLARs Tg mice, we generated a Reg3b?/? strain. Reg3b?/? mice grew to adulthood without apparent abnormalities. Deletion of Reg3b in CFLARs Tg mice exacerbated the embryonic lethality of CFLARs Tg mice. Dextran sulfate sodium-induced colitis, characterized by body weight loss and infiltration of neutrophils, was exacerbated in Reg3b?/? compared to wild-type mice. Moreover, the expression of Interleukin 6, an inflammatory cytokine and Chitinase-like 3, a marker for tissue repair macrophages was elevated in the colon of Reg3b?/? mice compared to wild-type mice after DSS treatment. Together, these results suggest that attenuation of colitis and ileitis is a result of Reg3β′s real function. 相似文献
Yamasinaium noduligerum is a Japanese fungivorous millipede that is usually found on decaying logs fallen on the forest floor. Here we present observations on paternal egg-brooding behavior in this species. At the end of May to early June, we found males with eggs on the bark of the underside of fallen decaying logs and in old burrows and galleries excavated by wood-dwelling insects under the bark of the logs. The males showed a typical brooding posture where they curl the anterior part of the body around the egg mass. The egg masses contained, on average, 30.2 ± 13.7 (SD) eggs. The brooding males showed no aggressive responses when disturbed. 相似文献
A PCR-mediated direct cloning for target spot DNA from RLGSgel has been established. The method consists of PCR amplificationof adaptor-ligated spot DNA fragments without excluding similar-sizedDNA fragments co-localized on RLGS gel, and following selectiveligation with the NotI-dT vector. Applying this method, we havesuccessfully cloned several DNA fragments derived from targetspots whose intensities change developmentally due to DNA methylationin the telencephalon of C3H/HeN mice. Since only a few microgramsof total DNA is sufficient for our spot cloning, our methodmay be highly useful when the total DNA sample prepared forcloning is limited. 相似文献

Abstract The CbpA protein is an analog of the DnaJ molecular chaperone of Escherichia coli . The dnaJ − cbpA − double-null mutant exhibits severe defects in cell growth, namely, a very narrow temperature range for growth. To gain insight into the functions of CbpA as well as DnaJ, we isolated a multicopy suppressor gene that permits this dnaJ − cbpA −~ mutant to grow normally at low temperatures. The suppressor gene was identified as rpoD , the gene that encodes the major σ 70. The biological implications of this finding are examined and discussed. 相似文献
Sixty-seven isolates of the southern blight fungus from Japan were divided into five groups based on ITS-RFLP analysis of
nuclear rDNA. Morphological characters of sclerotia varied between groups. Three groups were reidentified asSclerotium rolfsii, and two resembledS. delphinii in RFLP patterns and/or in having large sclerotia and relatively low optimal growth temperature (28°C). Sclerotia of the
latter, however, varied in size according to temperature and became indistinguishable from those ofS. rolfsii at high temperatures. Hyphal anastomosis (imperfect fusion) was observed between different ITS-RFLP groups, as well as between
different isolates belonging to the same groups. These results indicate that populations of this fungus in Japan consists

We report the case of an 84-year-old female with symptomatic bradycardia due to a complete atrioventricular block, who carried absent right and persistent left superior vena cava (SVC). Implantation of a pacing lead, particularly within the right ventricle (RV) in a patient with this venous anomaly is accompanied by technical difficulties. However, the apparatus consisting of a fixed-curve sheath (Model C315-S10, Medtronic, Inc., Minneapolis, MN, USA) and a lumenless fixed-screw pacing lead (Model 3830, Medtronic), allowed a rapid delivery into the RV without any complications. By rotating the Model C315-S10 sheath in the counterclockwise direction in the right atrium, its tip faced the tricuspid orifice, advanced across the tricuspid valve and confronted the RV lower septum near the apex. Then the RV-lead was fixed with acceptable pacing and sensing parameters. Utilizing a lumenless pacing lead and a preformed sheath to deliver it is a novel approach that could be helpful in pacemaker implantation in patients with absent right and persistent left SVC. 相似文献
